5 твердофазный синтез
метод синтеза полимеров (см. полимеры) и олигомеров (см. олигомеры) с использованием твердого нерастворимого носителя, представляющего собой органический или неорганический полимер. Т.с. основан на том, что первое звено будущего олигомера ковалентно закрепляется на "якорной" группе носителя, наращивание цепи проводится стандартно защищенными мономерами по обычным схемам, используемым для синтеза в растворах. На заключительном этапе синтезируемый олигомер отщепляется от носителя и очищается соответствующими методами. Т.с. применяют в основном для получения полипептидов, олигонуклеотидов и олигосахаридов. При синтезе полипептидов в качестве носителя часто используют сополимер стирола и 1—2 % дивинилбензола, модифицированный введением диметоксибензилхлоридной якорной группы для присоединения первой аминокислоты (с защищенной группой NH2) по С-концу. После удаления N-защитной группы наращивание полипептидной цепи проводят стандартными методами пептидного синтеза в растворе. В качестве конденсирующих агентов наиболее часто используют карбодиимиды или предварительно превращают аминокислоты в активированные эфиры. Т.с. предложен и введен в практику Р. Меррифилдом в 1962 г. для синтеза полипептидов (Нобелевская премия за 1984 г.), а затем распространен на синтез олигонуклеотидов (Р. Летзингер, 1964 г.) и олигосахаридов (А.
